Indications

A medication called Neurolin 50 is used to treat neuropathic pain, which is pain that develops as a result of nerve damage brought on by diabetes, shingles (herpes zoster infection), spinal cord injury, or other conditions. Additionally, it is used to treat widespread muscle pain and stiffness in fibromyalgia patients. In conjunction with other medications, Neurolin 50, a member of the anti-epileptic drug class, may be used to treat some types of seizures (fits). If other medications are ineffective, they may also be prescribed to treat the symptoms of excessive anxiety in generalized anxiety disorder.

Pharmacology

Gamma-aminobutyric acid (GAB) is an inhibitory neurotransmitter that may have a basic subsidiary in pregabalin (GABA). It is not directly linked to benzodiazepine, GABAA, or GABAB receptors. Pregabalin has a strong affinity for the central nervous system tissues that contain the assistant subunit of voltage-gated calcium channels, alpha-2-delta. Pregabalin has a 90% verbal bioavailability that is independent of measurements. It is excreted from the body primarily through the kidneys as a pure tranquilizer with a cruel end half-life of 6.3 hours in subjects with typical renal function.

Dosage & Administration

  • The maximum advised dose of Pregabalin (Neurolin?) for neuropathic pain related to diabetic peripheral neuropathy is 100 mg administered three times daily (300 mg/day). The dosage should start at 50 mg three times per day (150 mg/day), and depending on effectiveness and tolerability, it may be increased to 300 mg/day within a week.
  • Pregabalin (Neurolin) is prescribed in doses ranging from 75 to 150 mg twice daily or 50 to 100 mg three times daily (150 to 300 mg/day) for post-herpetic neuralgia. The dosage should start at 75 mg twice daily or 50 mg three times daily (150 mg/day), and depending on effectiveness and tolerability, it may be increased to 300 mg/day within a week.
  • Management of Fibromyalgia: The recommended dose of Pregabalin for fibromyalgia is 300 to 450 mg/day. Dosing should begin at 75 mg two times a day (150 mg/day) and may be increased to 150 mg two times a day (300 mg/day).
  • Pregabalin (Neurolin) has been shown to be effective as adjunctive therapy in the treatment of partial-onset seizures in adult patients when administered at doses of 150 to 600 mg/day. It is recommended to divide the recommended daily dose into two or three doses.
  • The dosage range for neuropathic pain brought on by spinal cord damage is 150 to 600 mg per day. The starting dosage is 75 mg twice daily, or 150 mg per day. Depending on effectiveness and tolerability, the dose may be increased within a week to 150 mg twice daily (300 mg/day).

Interaction

There is no noteworthy intelligent between Pregabalin with other antiepileptic drugs & verbal prophylactic. Pregabalin may potentiate the impacts of ethanol and lorazepam.

Contraindications

Pregabalin (Neurolin?) is contraindicated in patients with known hypersensitivity to Pregabalin or any of its components.

Pregnancy & Lactation

Discontinuation of Pregabalin (Neurolin?) without tapering may produce insomnia, nausea, headache, and diarrhea. So it should be tapered gradually over a minimum of 1 week rather than discontinued abruptly. Creatinine kinase may be elevated if treated with Pregabalin. It should be discontinued rapidly if myopathy is diagnosed or suspected or if creatinine kinase is elevated markedly.

Precautions & Warnings

Pregnancy: Pregnancy category C. So it should only be used if the potential benefit justifies the potential risks to the fetus.

Nursing mother: It is not known if pregabalin is excreted in human milk; it is, however, present in the milk of rats. So it should be used in nursing mothers only if there is a clear benefit over the risk.
