Indications of Tyclav 625mg Tablet

  • Tyclav 625mg Tablet is used for the short-term treatment of bacterial infections in the following locations:
  • Upper respiratory tract infections include tonsillitis, sinusitis, and otitis media (including ENT).
  • Lower respiratory tract infections include acute and chronic bronchitis, lobar pneumonia, and bronchopneumonia.
  • Cystitis, urethritis, and pyelonephritis are genito-urinary tract infections.
  • Infections of the skin and soft tissues.
  • Osteomyelitis is an infection of the bones and joints.
  • Septic abortion, puerperal sepsis, intra-abdominal sepsis, and other infections are all possibilities.

Pharmacology of Tyclav 625mg Tablet

Pharmacodynamic properties: 

  • Tyclav 625mg Tablet is an antibacterial combination of Amoxicillin and the (3-lactamase inhibitor Clavulanic Corrosive. Amoxicillin has a broad spectrum of bactericidal activity against many Gram-positive and Gram-negative microorganisms, but it is susceptible to degradation by (3-lactamases, and thus its range of action excludes microorganisms that produce these proteins. Clavulanic corrosive can inactivate a wide range of beta-lactamase proteins found in microorganisms resistant to penicillins and cephalosporins. As a result of this arrangement, Clavulanic corrosive protects Amoxicillin from degradation by (3-lactamase chemicals and successfully broadens the anti-microbial range to capture a wide range of microorganisms.

Pharmacokinetic properties: 

  • The pharmacokinetics of Tyclav 625mg Tablet’s two components are closely coordinated. The highest serum levels of both occur approximately one hour after verbal organization. Tyclav 625mg Tablet absorption is maximized at the start of a feast. Both clavulanate and amoxicillin have low serum levels; nearly 70% remain free in the serum. The serum levels were achieved by multiplying the dose of Tyclav 625mg Tablet by two.

Dose of Tyclav 625mg Tablet 

Adults and children over 12 years: 

Tablet:

  • Regular adult measurements are one 625 mg Tablet every 12 hours or one 375 mg Tablet every 8 hours. For more severe diseases and contaminations of the respiratory tract, one 1 gm Tablet every 12 hours or one 625 mg Tablet every 8 hours should be taken.

Suspension: 

  • For children aged 6 to 12: 2 teaspoonful every 8 hours. 1 teaspoonful every 8 hours for children aged 1-6 years.
  • Children under 1 year: 25 mg/kg/day in divided doses every 8 hours; for example, a 7.5 kg child would require 2 ml suspension t.i.d. Treatment should not be extended beyond 14 days without review.

Forte suspension: 

  • The standard prescribed daily measurement is 25/3.6 mg/kg/day in mild to severe contaminations (upper respiratory tract contaminations, for example, repetitive tonsillitis, lower respiratory contaminations, and skin and delicate tissue infections).
  • 45/6.4 mg/kg/day for the treatment of more serious diseases (upper respiratory tract contaminations such as otitis media and sinusitis, lower respiratory diseases such as bronchopneumonia, and urinary tract infections).

Children of 2 to 12 years: 

Mild to direct infections: 

  • 25/3.6 mg/kg per day (Suspension)
  • 2-6 years (13-21 kg) 2.5 mL b.i.d. suspension
  • 7-12 years (22-40 kg) 5 ml suspension b.i.d.

Adults- 

  • 1.2 gm every 8 hours is typical.
  • Increased in genuine contaminations to 1.2 gm every 6 hours
  • For surgical prophylaxis, the standard measurement is 1.2 gm at acceptance; however, for high-risk methods (e.g., colorectal surgery), up to 2-3, gm may be given every 8 hours.

Children-

  • 30 mg/kg every 8 hours for the first 3 months. (every 12 hours during the perinatal period and in premature infants.
  • 3 months to 12 years: 30 mg/kg every 8 hours, increased to 30 mg/kg every 6 hours in more severe disease.

Administration of Tyclav 625mg Tablet

Oral dose shape: 

  • This can be taken without regard for dinners; in any case, clavulanate potassium retention is improved when Amoxicillin and Clavulanic corrosive are administered at the start of dinner. Amoxicillin and Clavulanic corrosive should be taken at the start of the meal to reduce the risk of gastrointestinal narrow-mindedness.
  • IV infusion is not recommended for intramuscular or subcutaneous organizations. The reconstituted vial can be administered intravenously through infusion (lasting more than 2 minutes) or moderate intravenous implantation (30 minutes). The vial’s substance must be used within 20 minutes, after which any unused fabric must be disposed of.

Interaction of Tyclav 625mg Tablet

A few patients who took Tyclav 625mg Tablet had their dying time and prothrombin time extended. Tyclav 625mg Tablet, like other broad-spectrum antimicrobials, may reduce the efficacy of oral contraceptives, and patients should be warned accordingly. Concurrent use of allopurinol with amoxicillin can increase the likelihood of unfavorably susceptible skin reactions. There is no information on the use of Tyclav 625mg Tablet and allopurinol together.

Contraindications of Tyclav 625mg Tablet

History of Penicillin extreme touchiness. Consideration ought to be paid to conceivable cross-sensitivity with other beta-lactam antibiotics e.g. cephalosporins. Moreover contraindicated for patients with a past history of Tyclav 625mg Tablet or Penicillin-associated cholestatic jaundice.

Tyclav 625mg Tablet Side effects

  • As with Amoxicillin, side effects are uncommon and mostly mild and transient in nature. 
  • Loose bowels, pseudomembranous colitis, acid reflux, sickness, heaving, and candidiasis have all been reported. 
  • If gastrointestinal side effects occur with verbal treatment, they can be reduced by taking Tyclav 625mg Tablet at the start of meals. 
  • Hepatitis and cholestatic jaundice have been mentioned on occasion, but they are usually reversible.
  • In some cases, urticarial and erythematous rashes develop. 
  • Occasionally, erythema multiforme, Stevens-Johnson syndrome, and exfoliative dermatitis have been discussed. 
  • Angioedema and anaphylaxis have been described, as have other beta-lactam antimicrobials.

Pregnancy & Lactation

Tyclav 625mg Tablet has been demonstrated in animal experiments to have no teratogenic effects when given orally or parenterally. The drug has been taken orally in human pregnancy in a small number of cases with no adverse effects; nonetheless, it is not recommended to take Tyclav 625mg Tablet during pregnancy unless it is declared necessary by your doctor. Amoxicillin can be found in trace amounts in breast milk during lactation.

Precautions & Warnings

  • Tyclav 625mg Tablet should be used with caution in patients taking anticoagulant medication or who have significant hepatic impairment.
  • In patients with moderate or severe renal impairment, the dosage should be reduced.
  • To avoid crystalluria, maintain adequate fluid intake and urine output during the administration of a high dose of Tyclav 625mg Tablet.

Storage Conditions

This should be kept at a temperature below 25?C, away from light and moisture. Suspension should be kept refrigerated (not frozen) and used within 7 days of being reconstituted. The vial must be used within 20 minutes of being reconstituted.
